چکیده

Intracytoplasmic membrane vesicles (chromatophores) isolated from Rhodopseudomonas capsulata cells were fused with liposomes by a pH transition procedure. Vesicles o f lower density and higher lipid contents and larger diameter than chromatophores were obtained. Similar results were observed by Ca2+ induced fusion and by the freeze-thawing method. Respiratory and light-induced electron transport were measured in chromatophores and fused vesicles. Light-induced reaction center bleaching was observed in all types o f vesicles, whereas repiratory electron transport was substantially diminished by lipid incorporation. Ubiquinone 10 restored to some extent respiratory electron transport and oxidative phosphorylation and it m odified the photophosphorylation kinetics under continuous light. Electrochromic carotenoid band-shift and the 9-aminoacridine fluorescence quenching indicate that the capacity o f the fused vesicles to maintain an electrochemical proton gradient has not been substantially diminished. From the kinetics of 9-aminoacridine quenching an increased K+-permeability seems to be apparent.

Engraftment of plasma membrane vesicles into liposomes: A new method for designing of liposome-based vaccines

Objective(s):One of the major challenges in the field of vaccine design is choosing immunogenic antigens which can induce a proper immune response against complex targets like malignant cells or recondite diseases caused by protozoan parasites such as leishmaniasis. The aim of this study was to find a way to construct artificial liposome-based cells containing fragments of target’s cell membran...

Membrane fusion of secretory vesicles and liposomes. Two different types of fusion.

Secretory vesieles isolated from adrenal medulla were found to fuse in vitro in response to incubation with Ca2+. Intervesicular fusion was detected by electron microscopy and was indicated by the appearance of twinned vesieies in freeze-fractured suspensions of vesieles and in thin-sectioned pellet. Two types of fusion could be distinguished: Type I, occurring between 10M and 10M Ca+, was spec...

Calcium-dependent fusion of the plasma membrane fraction from human neutrophils with liposomes.

A cell-free assay monitoring lipid mixing was used to investigate the role of Ca2+ in neutrophil membrane-liposome fusion. Micromolar concentrations of Ca2+ were found to directly stimulate fusion of inside-out neutrophil plasma membrane enriched fractions (from neutrophils subjected to nitrogen cavitation) with liposomes (phosphatidylethanolamine:phosphatidic acid, 4:1 molar ratio). In contras...

Actual fusion efficiency in the lipid mixing assay - Comparison between nanodiscs and liposomes

Lipid exchange occurs between membranes during fusion or active lipid transfer. These processes are necessary in vivo for the homeostasis of the cell at the level of the membranes, the organelles and the cell itself. They are also used by the cell to interact with the surrounding medium. Several assays have been developed to characterize in vitro these processes on model systems. The most commo...

Fusion between fluid liposomes and intact bacteria: study of driving parameters and in vitro bactericidal efficacy

BACKGROUND Pseudomonas aeruginosa represents a good model of antibiotic resistance. These organisms have an outer membrane with a low level of permeability to drugs that is often combined with multidrug efflux pumps, enzymatic inactivation of the drug, or alteration of its molecular target. The acute and growing problem of antibiotic resistance of bacteria to conventional antibiotics made it im...
